SATB2-associated Syndrome

SATB2-associated Syndrome is a multisystem neurodevelopmental disorder characterized by moderate-to-profound developmental impairment, absent or severely limited speech, palatal and dental abnormalities, feeding difficulty, hypotonia and behavioral features. Epilepsy, low bone density and craniofacial abnormalities are variable.

Genetics

Caused by a heterozygous intragenic SATB2 pathogenic variant, deletion involving SATB2 or chromosomal rearrangement disrupting the locus. Most cases are de novo and represent autosomal dominant haploinsufficiency.

Molecular Mechanism

SATB2 is a chromatin organizer and transcriptional regulator required for cortical projection-neuron identity, craniofacial patterning and osteoblast differentiation. Reduced dosage disrupts coordinated gene-expression programmes in brain, palate, teeth and bone.

Therapy

Treatment is supportive, with early augmentative communication, feeding and developmental therapy, dental and palatal management, seizure care and monitoring of growth and bone density. No disease-modifying therapy is established.

CCP Activity

CCP is developing inducible SATB2 expression restoration model.
