AGO1-related Neurodevelopmental Disorder

AGO1-related Neurodevelopmental Disorder is a recently defined neurodevelopmental disorder with variable intellectual disability, severe speech and language impairment, hypotonia, motor delay, behavioral or autistic features and, in a subset, epilepsy and brain abnormalities.

Genetics

Usually caused by de novo heterozygous coding variants in AGO1. Pathogenic missense variants cluster in conserved functional domains and cannot be interpreted simply as AGO1 haploinsufficiency.

Molecular Mechanism

AGO1 is a core component of the microRNA-induced silencing complex. Disease variants alter microRNA loading, target recognition or silencing dynamics and can interfere with the function of the normal allele, producing broad post-transcriptional dysregulation during neurodevelopment.

Therapy

Treatment is supportive and directed to epilepsy, motor impairment, communication, feeding, behavior and sleep. No established molecular therapy is available; functional classification of individual variants is a prerequisite for future allele-specific strategies.

CCP Activity

CCP is developing and characterizing an AGO1 disease model. The programme links systemic and neurobehavioral phenotyping.
