Czech Centre for Phenogenomics at the Czech Science Fair / České centrum pro fenogenomiku na Veletrhu vědu

We are from the Czech Centre for Phenogenomics at the Institute of Molecular Genetics of the CAS, located in the outskirts of Prague in Vestec in the BIOCEV complex. And what exactly does phenogenomics mean and what do we do? Phenogenomics is a field that combines genomic analysis and phenotype of an organism. It investigates how a change in gene function affects the physical and biochemical properties of an organism. In our centre, we analyse mice in which we cut out/deactivate a gene and use comprehensive tests to determine how the deletion of this gene affects the mouse. The mouse undergoes neurobehavioral tests, and we also determine the functionality of the lungs, heart, eyes and hearing. We’ll measure its metabolism and response to glucose. We use the CRISPR-Cas9 system to create our mouse models. This system is also known as biological scissors. We will design 2 short RNAs (CRISPR) that will find a spot on the genomic DNA at the site of the gene we want to cut. Then the Cas9 enzyme will cleave the DNA. The repair mechanism, which works in every cell, then joins the ends of the genomic DNA together and we’re done. To ensure that the resulting genetically modified mouse lacks our designated gene in all cells, the gene is excised at the 2-4 cell stages of the mouse embryo.

Jsme z Českého centra pro fenogenomiku při Ústavu molekulární genetiky AV ČR, které se nachází na okraji Prahy ve Vestci v komplexu BIOCEV. A co vlastně znamená ta fenogenomika a čím se tedy zabýváme? Fenogenomika je obor, který kombinuje genomickou analýzu a fenotyp organismu. Zjišťuje, jak se projeví změna funkce genu na fyzikálních a biochemických vlastnostech organismu. V našem centru mimo jiné analyzujeme myši, kterým vystřihneme/deaktivujeme jeden gen a pomocí komplexních vyšetření zjišťujeme, jak se projeví delece tohoto genu na myši. Myš projde neurobehaviorálními testy, dále zjišťujeme funkčnost plic, srdce, očí a sluchu. Změříme její metabolismus a reakci na glukózu. Pro vytvoření našich myších modelů používáme CRISPR-Cas9 systém. Tomuto systému se také přezdívá biologické nůžky. Navrhneme 2 krátké RNA (CRISPR), které najdou místo na genomové DNA v místě genu, který chceme vystřihnout. Pak už jen enzym Cas9 rozštěpí DNA. Opravný mechanismus, který funguje v každé buňce, k sobě poté spojí konce genomové DNA a je hotovo. Aby vzniklé geneticky modifikované myši chyběl námi určený gen ve všech buňkách, dochází k vystřižení genu na 2-4 buněčných stádiích myšího embrya.
